Glycolic Acid for Single-Step Cleaning and Disinfection in the Food Industry: Efficiency, Safety, and Sustainability

Disinfectant mechanism of action of glycolic acid

Glycolic acid is included in the Article 95 list of the Biocidal Regulation (EU) No 528/2012 as a biocidal active substance for product types PT2, PT3, and PT4

The disinfectant action of glycolic acid is based on the combination of the following mechanisms of action on microorganisms:

  1. pH alteration:
    • Glycolic acid lowers the pH of the medium, as it is a weak acid with pKa ≈ 3.8).
    • Many microorganisms (bacteria, fungi) do not tolerate highly acidic environments.
  1. Protein denaturation:
    • At low pH, the acid induces protonation of functional groups in bacterial proteins, altering their structure and inactivating enzymes essential for microbial survival.
  1. Cell membrane disruption:
    • Glycolic acid easily penetrates membranes due to its small and polar molecular structure.
    • Inside the cell, it acidifies the cytoplasm and disrupts the proton gradient, potentially leading to cell lysis or bacterial death.
  1. Synergistic effect with other agents:
    • In disinfectant formulations, glycolic acid can enhance the action of other organic ingredients, helping to break down biofilms and improve penetration.
